Measuring hundreds of proteins at once with precision seems impossible: antibodies get confused and false signals appear. PEA technology (Proximity Extension Assay) solves exactly that. It is the heart of Olink and the reason its data is so clean.
Each protein is recognized by a pair of antibodies, and each antibody carries a small DNA tag. When both antibodies bind the same protein, their tags end up so close that they join and form a new piece of DNA. That piece is like a unique barcode for that protein: it is copied, read and counted. The more protein there is, the more barcode signal is generated.
Each protein needs two antibodies to be right at the same time. If only one is wrong, there is no signal. That is why specificity is very high and false positives minimal.
By turning each protein into a unique DNA tag, hundreds or thousands can be measured at once in the same sample without interfering. That is the secret behind the high multiplexing.
The DNA readout is amplified, so it can detect even very low-abundance proteins from just a few microliters of blood.
